Resumen

Esta investigación tuvo como objetivo principal realizar el aislamiento de la Salmonella Gallinarum (Tifoidea aviar) y de la Salmonella Pullorum (Pulorosis) en una planta de sacrificio, mediante la utilización de hisopos cloacales en las aves, luego el aislamiento de los microorganismos en medios de cultivo específicos y su posterior identificación empleando pruebas bioquímicas, así como el equipo para la identificación de patógenos entéricos no fermentadores (BBL CRYSTAL) y de esta manera determinar su prevalencia en esta etapa de sacrificio del pollo. Se Implementó el manual POE de VENEGAS, SANCHES, GOMEZ Y SANTOS, 2009, El cual contribuye para obtener una base informativa que facilite las políticas de identificación y control de salmonelosis en el país propuestas por el COMPES 3486, La selección de las muestras se realizó completamente al azar hasta completar el número de aves totales por muestreo; se trabajaron 4 lotes de pollo de engorde de la línea Ross 308, de los cuales se obtuvo 20 muestras de cada lote, cabe recordar que cada muestra equivale a 20 aves, durante 3 semanas para un total de 240 muestras que corresponden a un total de 48.000 aves muestreadas (4x20x3); es de aclarar que para su posterior análisis se utilizó estadística descriptiva. Las muestras tomadas por medio de hisopos cloacales fueron sumergidas en tubos con agua peptonada al 5% y llevadas a 37˚C por 12 horas; luego de retirar el hisopo se tomo 1 ml de cada muestra y se colocó en tubos con caldo tetrationato a 37˚C por 12 horas. Posteriormente mediante un asa estéril se sembró con la técnica de agotamiento en medio de cultivo XLD y se llevó a una temperatura de 37˚C por 12 horas. Transcurrido este tiempo fueron seleccionadas las colonias con características de cambio de color en el medio propias de la Salmonella y se realizaron pruebas bioquímicas (catalasa, oxidasa, indo y movilidad). Las muestras consideradas como positivas a Salmonella se sembraron en agar Tripticasa soya a 37˚C por 24 horas, para luego realizarles pruebas con el Kid Diagnosticó BBL CRYSTAL. Del total de 240 muestras analizadas, se encontró que en 185 se observaron características positivas para Salmonella, las cuales presentaron colonias con pigmentación roja o rosa y en algunos casos según el manual Merck colonias trasparentes con o sin centro negro o completamente negras para un total del 77%. Las 55 muestras restantes se descartaron ya que no cumplieron con las especificaciones de las colonias de Salmonella y correspondieron al 23% del total. En las pruebas realizadas con BBL CRYSTAL se encontró Salmonella Spp y otros agentes patógenos que se detectaron en la presente investigación como fueron el Enterobacter cloacae 20%, Pseudomona stutzeri 10% y la Klebsiella pneumoniae 10%. Como las serovariedades Salmonella Gallinarum y Salmonella Pullorum no producen H2S, y en nuestros resultados las salmonellas si produjeron H2S, consideramos innecesario realizar las pruebas de serotipificación para confirmar la presencia de dichas serovariedades. De acuerdo a los resultados obtenidos en este trabajo y aunque en principio fueron aisladas Salmonellas Spp, en las muestras trabajadas, no se confirma la presencia de las serovariedades objeto principal del estudio como son la Salmonella Pullorum y la Salmonella Gallinarum.

This investigation had as principal aim realize the isolation of the Salmonella Gallinarum and of the Salmonella Pullorum (Pulorosis) in a plant of sacrifice, by means of the utilization of hyssops cloacale in the birds, then the isolation of the microorganisms in specific means of culturing and his later posterior identification using biochemical tests, as well as the equipment for the identification of pathogenic enteric not fermentation (BBL CRYSTAL) and hereby to determine his prevalence in this stage of sacrifice of the chicken. There was Implemented the manual POE of Venegas, Sanches, Gomez And Saints, 2009, Which contributes(pays) to obtain an informative base that facilitates the policies of identification and control of Salmonelosis in the country proposed by the COMPES 3486, The selection of the samples was realized completely at random up to completing the number of total birds for sampling; they worked 4 lots of chicken of there puts on weight of the line Ross 308, of which there were obtained 20 samples of every lot, it is necessary to remember that every sample is equivalent to 20 birds, for 3 weeks for a total of 240 samples that correspond to a total of 48.000 sampled birds (4x20x3); it is of clarifying that for his later analysis descriptive statistics was in use. The samples taken by means of hyssops cloacale were immersed in pipes by water peptonada to 5 % and rides to 37°C for 12 hours; after withdrawing the hyssop I take 1 ml of every sample and he(she) placed in pipes with broth tetrationato to 37°C for 12 hours. Later by means of a sterile handle sowed with the technology (skill) of depletion in the middle of culture (culturing) XLD and led to a temperature of 37°C for 12 hours. Passed this time the colonies were selected by characteristics of change of color in the way own of the Salmonella and there were realized biochemical tests (catalasa, oxidasa, Indus and mobility). The samples considered since positive to Salmonella Tripticasa sowed in agar soy bean to 37°C for 24 hours, then to realize tests with the Kid Diagnostic BBL CRYSTAL. Of the total of 240 analyzed samples, one thought that in 185 positive characteristics were observed for Salmonella, which presented colonies with red or pink pigmentation and in some cases according to the manual Merck colonies transparences with or without black center or completely black for a total of 77 %. 55 remaining samples discarded since they did not expire with the specifications of Salmonella's colonies and corresponded to 23 % of the total. In the tests realized with BBL CRYSTAL one found Salmonella Spp and other pathogenic agents who detected to themselves in the present research Since they were the Enterobacter cloacae 20 %, Pseudomona stutzeri 10 % and the Klebsiellap neumoniae 10 %. As the serovariedades Salmonella Gallinarum and Salmonella Pullorum do not produce H2S, and in our results the salmonellas if they produced H2S, we think unnecessarily to realize the tests of serotipificatión to confirm the presence of said serovariedades. In agreement to the results obtained in this work and though at in principle)Salmonella´s Spp was isolated, in the worn out samples, the presence of the serovariedades does not confirm to itself principal object of the study they are Salmonella Pullorum and the Salmonella Gallinarum.
